Test developed to detect early-stage diseases with naked eye: Prototype ultra sensitive disease sensor developed

ScienceDaily (Oct. 28, 2012) ? Scientists have developed a prototype ultra-sensitive sensor that would enable doctors to detect the early stages of diseases and viruses with the naked eye, according to research published?October 28 in the journal Nature Nanotechnology.

The team, from Imperial College London, report that their visual sensor technology is ten times more sensitive than the current gold standard methods for measuring biomarkers. These indicate the onset of diseases such as prostate cancer and infection by viruses including HIV.

The researchers say their sensor would benefit countries where sophisticated detection equipment is scarce, enabling cheaper and simpler detection and treatments for patients.

In the study, the team tested the effectiveness of the sensor by detecting a biomarker called p24 in blood samples, which indicates HIV infection.

Professor Molly Stevens, from the Departments of Materials and Bioengineering at Imperial College London, says: "It is vital that patients get periodically tested in order to assess the success of retroviral therapies and check for new cases of infection. Unfortunately, the existing gold standard detection methods can be too expensive to be implemented in parts of the world where resources are scarce. Our approach affords for improved sensitivity, does not require sophisticated instrumentation and it is ten times cheaper, which could allow more tests to be performed for better screening of many diseases."

The researchers in this study also tested samples for the biomarker called Prostate Specific Antigen (PSA), which is an early indicator for Prostate Cancer. The team say the sensor can also be reconfigured for other viruses and diseases where the specific biomarker is known.

The sensor works by analysing serum, derived from blood, in a disposable container. If the result is positive for p24 or PSA, there is a reaction that generates irregular clumps of nanoparticles, which give off a distinctive blue hue in a solution inside the container. If the results are negative the nanoparticles separate into ball-like shapes, creating a reddish hue. Both reactions can be easily seen by the naked eye.

The team also report that the sensor was so sensitive that it was able to detect minute levels of p24 in samples where patients had low viral loads, which could not be diagnosed using existing tests such as the Enzyme-linked Immunosorbent Assay (ELISA) test and the gold standard nucleic acid based test.

Dr Roberto de la Rica, co-author of the study from the Department of Materials at Imperial College London, adds: "We have developed a test that we hope will enable previously undetectable HIV infections and indicators of cancer to be picked up, which would mean people could be treated sooner. We also believe that this test could be significantly cheaper to administer, which could pave the way for more widespread use of HIV testing in poorer parts of the world."

The next stage of the research will see the team approaching not-for-profit global health organisations, which could provide strategic direction and funding for manufacturing and distributing the sensor to low income countries.

Full moon could make Sandy's impact worse

Heaven and Earth may be aligning to turn Hurricane Sandy into a real monster, just in time for Halloween.

Forecasters expect Sandy to make landfall along the mid-Atlantic coast of the United States on Monday or Tuesday. It may merge with a separate tempest hitting the region at about that time, creating an immensely powerful "Frankenstorm" whose effects could be magnified by the full moon.

When the moon waxes to its full phase Monday afternoon, high tides along the Eastern Seaboard will rise about 20 percent higher than normal, even without the help of Sandy's storm surge, said Joe Rao, a meteorologist for News 12 in Westchester, N.Y. (Storm surges occur when a hurricane's winds push the water surface above normal levels.)

"Add a big storm, and the full moon will only serve to exacerbate the situation by helping to accentuate the higher-than-normal water levels, possibly leading to moderate or major coastal flooding and beach erosion," Rao told OurAmazingPlanet via email.

A gravitational lag usually causes the highest tides to come a day or two after every full moon, he added.

"So the worst coastal flooding wrought by Sandy might actually come on Tuesday, all depending on exactly where and when the storm makes landfall," said Rao, who is also the Night Sky Columnist for OurAmazingPlanet sister site SPACE.com. [ How to Prepare for Hurricane Sandy ]

At full moon, the Earth, sun and moon are arranged in a line, with Earth in the middle. Tidal ranges are especially high at this time because the gravitational tugs of the sun and moon on our planet reinforce each other. The same effect is felt at new moon, when the three bodies all line up, with the moon between Earth and the sun.

Indeed, tides at the new and full moons are called "spring tides." The term has nothing to do with the season; it comes from the German verb "springen," which means "to spring up," Rao said.

Full and new moons have magnified storm impacts in the past, such as during the devastating "Ash Wednesday" tempest that rocked the U.S. East Coast in March 1962. The moon was full when this storm hit, but it was also at perigee ? the point in its orbit when it comes closest to Earth ? making things even worse.

A close lunar approach is one thing folks in Sandy's path don't have to worry about ? small comfort, perhaps, but it's something.

"If there is any saving grace in this upcoming situation regarding Sandy, it is that Monday's full moon comes less than three days before it arrives at apogee (farthest from the Earth)," Rao said.

Deus Ex is the greatest PC game of all time. You might disagree, and if you do I forgive you, but there's a reason ION Storm's first-person conspiracy stealth role-playing game shooter routinely tops lists of the greatest PC games of all time. It combined so many different elements of genres into a single, compelling first-person experience with a fantastic story, and let players decide exactly how they want to play the game not just with moral choices, but strategic ones. You could be a stealthy assassin, a pacifist, a hacker, a commando, or any other type of character, and each role would have a way for you to play. Arkane Studios and Bethesda Softworks' Dishonored for the PC, Xbox 360?, and PlayStation 3? taps into the feeling and produces a compelling experience that lets you feel as brutal or subtle, malicious or passive as you want to be. Dishonored, available for $59.99 (list), feels like a spiritual successor to Deus Ex in the same way that Bioshock was a spiritual successor to System Shock (but without the shared development team; Warren Spector and the old ION Storm team had nothing to do with this game).

A Different London
Dishonored takes place in the city of Dunwall, a Victorian London-style capital of a Victorian England-style nation filled with fantastic, whale oil-powered technology that feels much more like a science fiction penny dreadful than the catch-all genre of "steampunk." A plague is spreading through the city, turning bystanders into monsters and rats into packs of man-eating street piranha. You play Corvo Attano, the Empress' bodyguard who returns to Dunwall after looking for aid for the city only to see the Empress killed before him. He's arrested for the crime and is scheduled to be executed, but he's broken out by loyalists to the Empress and her missing daughter who want to take down the brutal dictatorship that rose in her place. There's also a mysterious dark god who gives you magic powers and is very interested in your quest for revenge.

The game is first-person, but like Deus Ex it's not a run-and-gun shooter. Corvo is a powerful fighter with sword, pistol, and crossbow, and he only becomes more powerful as he upgrades his equipment and powers by collecting treasure and hidden runes and charms, but he's not invincible. With enough ammunition and preparation you can cut a swath through guards and decimate everyone in your path, but that's only one way to get through the game, and it has its own consequences. Corvo's powers and equipment let him sneak along rooftops, teleport short distances, and detect guards through walls, so you can sneak past enemies without letting them know you're there and without killing any of them. You can explore areas and find alternate routes with different dangers but fewer guards. You can even work with Dunwall's criminal underworld, performing favors for access to paths you couldn't find otherwise.

These paths all have obstacles that themselves offer different solutions, depending on your play style. If you want to knock a few guards out with sleeping darts, you can. If you want to teleport behind them and cut their throats, you can. If you want to set razor-filled land mines in guards' paths and watch them get shredded before using the distraction to get past them, you can. You can also rewire security systems to get past deadly walls of light, summon swarms of rats to eat people, stop time and kill everyone in the room before they see you, and summon blasts of wind to knock them through windows and off balconies. Each of these methods feel unique and satisfying, and no matter how you build up Corvo you'll feel unstoppable in some way. Dishonored lets you choose how to play through the entire game, then give you a half dozen other ways to play again to give you a new experience getting to the same objective.

Different Ways to Play
How you play affects the rest of the game, as well. Everyone you kill and every guard you alert adds to the chaos of the city, which makes it darker and more plague-filled. Depending on who you help or kill in missions, different paths will be available to you. The game takes place in discrete, connected zones for each mission and there's no open-world exploration outside, but it makes the story seem much more focused and rich than the excellent Elder Scrolls games. You aren't an adventurer and you're not looking through every cave and ruin at your leisure. You're an assassin who creeps through a city that considers you a traitor. It adds much more direction to the story, while giving you just enough room to stretch out and find different solutions.

Dishonored isn't perfect, though. The levels tend to feel a bit cramped despite their multiple paths, and while the transition from stealth to combat feels more organic than Deus Ex: Human Revolution, it isn't quite as seamless as the original Deus Ex. It's also very easy to rely on Dark Vision to see enemies around corners, which means your Dishonored experience will be just as monochrome and missing the visually impressive surroundings as Batman: Arkham Asylum.

Dishonored is a fantastic game that puts you in a playground of murder and stealth while still keeping focused on an interesting story in a rich and enthralling world. It's not quite as large, as open, or as well-written as Deus Ex, but it stands as a solid spiritual successor to one of the best PC games of all time. It's Bioshock to Deus Ex's System Shock 2, and it's worth a look no matter your favorite genre.

(AP) ? The U.S. military did not quickly intervene during the attack on the U.S. Consulate in Libya last month because military leaders did not have adequate intelligence information and felt they should not put American forces at risk, Defense Secretary Leon Panetta said Thursday.

In his most extensive comments to date on the unfolding controversy surrounding the attack in Benghazi, Panetta said U.S. forces were on heightened alert because of the anniversary of 9/11 and prepared to respond. But, he said, the attack happened over a few hours and was over before the U.S. had the chance to know what was really occurring.

"(The) basic principle is that you don't deploy forces into harm's way without knowing what's going on; without having some real-time information about what's taking place," Panetta told Pentagon reporters. "And as a result of not having that kind of information, the commander who was on the ground in that area, Gen. Ham, Gen. Dempsey and I felt very strongly that we could not put forces at risk in that situation."

Panetta was referring to Gen. Carter Ham, the head of U.S. Africa Command, and Gen. Martin Dempsey, chairman of the Joint Chiefs of Staff.

In a letter to President Barack Obama on Thursday, House Speaker John Boehner questioned whether the White House considered military options during or immediately after the attack, and he questioned what the president knew about the security threats in the country. He said that the national debate over the incident shows that Americans are concerned and frustrated about the administration's response to the attack.

"Can you explain what options were presented to you or your staff, and why it appears assets were not allowed to be pre-positioned, let alone utilized? If these reports are accurate, the artificial constraint on the range of options at your disposal would be deeply troubling," Boehner, R-Ohio, wrote.

U.S. Ambassador Chris Stevens and three other Americans were killed in the attack, which has become a heated campaign issue less than two weeks before the election. Republicans have criticized the Obama administration's failure to more quickly acknowledge that intelligence suggested very early on that it was a planned terrorist attack, rather than spontaneous violence erupting out of protests over an anti-Muslim film.

House and Senate Republicans as well as GOP presidential candidate Mitt Romney have criticized President Barack Obama and administration officials over the response to the attack and whether officials failed to provide enough security at the consulate.

And there have been ongoing questions about whether there should have been additional military forces sent to the consulate immediately after it became clear that the Americans were under attack.

As the events were unfolding, the Pentagon began to move special operations forces from Europe to Sigonella Naval Air Station in Sicily. U.S. aircraft routinely fly in and out of Sigonella and there are also fighter jets based in Aviano, Italy. But while the U.S. military was at a heightened state of alert because of 9/11, there were no American forces poised and ready to move immediately into Benghazi when the attack began.

Also, the Pentagon would not send forces or aircraft into Libya ? a sovereign country ? without a request from the State Department and the knowledge or consent of the host nation.

During his news conference, Panetta lamented the "Monday morning quarterbacking" that has been going on about how the U.S. handled the attack. And Dempsey, sitting alongside Panetta, bristled at questions about what the military did or did not do in the aftermath.

Noting that there are reviews already going on, Dempsey added, "It's not helpful, in my view, to provide partial answers. I can tell you, however, sitting here today, that I feel confident that our forces were alert and responsive to what was a very fluid situation."

Military officials have noted that there had been no specific intelligence pointing to a possible attack in Libya, and that there were protests and other unrest in various locations around the Middle East and Africa.

Asked whether the military was on alert in that area because of 9/11, Dempsey added, "Yeah, and let me point out, it was ? it was 9/11 everywhere in the world."

The Senate Intelligence Committee has announced it will hold a closed hearing Nov. 15 into the circumstances surrounding the terrorist attack, including the intelligence and security situation there, and additional hearings will follow.

ScienceDaily (Oct. 25, 2012) ? Had a bad day? Extending your normal exercise routine by a few minutes may be the solution, according to Penn State researchers, who found that people's satisfaction with life was higher on days when they exercised more than usual.

"We found that people's satisfaction with life was directly impacted by their daily physical activity," said Jaclyn Maher, graduate student in kinesiology. "The findings reinforce the idea that physical activity is a health behavior with important consequences for daily well-being and should be considered when developing national policies to enhance satisfaction with life."

The team examined the influence of physical activity on satisfaction with life among emerging adults ages 18 to 25 years because this population's sense of well-being appears to worsen more quickly than at any other time during adulthood.

"Emerging adults are going through a lot of changes; they are leaving home for the first time and attending college or starting jobs," said Maher. "As a result, their satisfaction with life can plummet. We decided to focus on emerging adults because they stand to benefit the most from strategies to enhance satisfaction with life."

The researchers recruited two groups of college students at Penn State. The first group, consisting of 190 individuals, entered information into a diary every day for eight days. The second group, consisting of 63 individuals, entered information into a secure website every day for 14 days. Both groups answered questions aimed at determining participants' satisfaction with life, physical activity and self-esteem. The personalities of all participants in the first group were assessed at the outset of the study using the Big Five Inventory short form.

For the second group (the 63 individuals who filled out questionnaires online for 14 days), the researchers wanted to further investigate whether physical activity was indeed, the cause of participants' increased satisfaction with life rather than some other factor such as mental health, fatigue, or Body Mass Index.

"Shifts in depression, anxiety and stress would be expected to influence a person's satisfaction with life at any given point in time," said David Conroy, professor of kinesiology. "In addition, fatigue can be a barrier to engaging in physical activity, and a high Body Mass Index associated with being overweight may cause a person to be less satisfied in a variety of ways."

By controlling for these variables, the researchers were able to determine that the amount of physical activity a person undertakes in a particular day directly influences his or her satisfaction with life. Specifically, the team found that by exercising just a little more than usual a person can significantly improve his or her satisfaction with life.

The results appeared online this week in the journal Health Psychology.

"Based on these findings, we recommend that people exercise a little longer or a little harder than usual as a way to boost satisfaction with life," said Conroy.

The National Institute of Aging at the National Institutes of Health funded this research. Other authors on the paper include Shawna Doerksen, assistant professor of recreation, park and tourism management; Steriani Elavsky, assistant professor of kinesiology; Amanda Hyde, graduate student in kinesiology; Aaron Pincus, professor of psychology; and Nilam Ram, associate professor of human development and family studies and of psychology.
